Theophylline Riboswitch (Clone 8.1*)
Theophylline Riboswitch Clone 8.1* encodes a RNA secondary structure that acts as a translational activator. In the presence of theophylline, the theophylline aptamer binds it inducer and causes a conformational change in the RNA structure that makes available the previously sequestered RBS, thus allowing translation of the downstream sequence. This riboswitch provides a 59 - fold activation ratio in 1mM theophylline as reported by Gallivan and Lynch in the figures below [1].
We, the UC Davis iGEM 2013 team, used this riboswitch to control the expression of TAL repressors in our RiboTAL constructs, please see parts K1212011 and K1212012 for further characterization.
